چکیده

Background: The accumulation of calcium by mitochondria regulates cell survival. Results: Purified mitochondria exhibit increased calcium uptake in the presence of NMDA receptor agonists. Conclusion: Functional NMDA-like receptors may be present on mitochondria. Significance: Learning how NMDA facilitates mitochondrial calcium uptake provides a new dimension to the understanding of cellular homeostasis and response to noxious stimuli.

Alteration of mitochondrial calcium homeostasis by ammonia-induced activation of NMDA receptors in rat brain in vivo.

The aim of the present work was to assess the effects of activation of NMDA receptors in rat brain in vivo on calcium homeostasis in isolated non-synaptic brain mitochondria. We have shown recently that acute intoxication with large doses of ammonia leads to activation of NMDA receptors in rat brain in vivo. In the present work we injected rats with ammonium acetate to activate NMDA receptors i...

Privileged access to mitochondria of calcium influx through N-methyl-D-aspartate receptors.

Mitochondrial Ca2+ uptake responds dynamically and sensitively to changes in cytosolic Ca2+ levels and plays a crucial role in sequestering the large Ca2+ load induced by N-methyl-D-aspartate (NMDA) receptor activation. However, the precise interrelationships between NMDA receptor activation, cytosolic Ca2+ increase, and mitochondrial Ca2+ uptake remain obscure. To reliably, independently, and ...

4-hydroxy-2(E)-Nonenal facilitates NMDA-Induced Neurotoxicity via Triggering Mitochondrial Permeability Transition Pore Opening and Mitochondrial Calcium Overload

N-methyl-D-aspartate (NMDA) receptor-mediated excitotoxicity is one of the major causes for neuronal cell death during cerebral ischemic insult. Previously, we reported that the final product of lipid membrane peroxidation 4-hydroxy-2E-nonenal (HNE) synergistically increased NMDA receptor-mediated excitotoxicity (J Neurochem., 2006). In this study, we investigated the mechanism involved in the ...

Protein - mediated Uptake of Vitamin 12 by Isolated Mitochondria

Protein-mediated B12 uptake by isolated respiration. Following uptake, cyanorat liver mitochondria has been shown to cobalamin is converted to adenosyl and be enhanced by plasma transcobalamin methylcobalamins and released from the (TC-lI) but not by salivary R binder in mitochondria. TC-ll appears to be required vitro. The process is enhanced by calcium for both cellular and mitochondrial upta...

Protein-mediated uptake of vitamin B12 by isolated mitochondria.

Protein-mediated B12 uptake by isolated rat liver mitochondria has been shown to be enhanced by plasma transcobalamin (TC-II) but not by salivary R binder in vitro. The process is enhanced by calcium and depends on active mitochondrial respiration. Following uptake, cyanocobalamin is converted to adenosyl and methylcobalamins and released from the mitochondria. TC-II appears to be required for ...
